Multinuclear NMR studies of the interaction of metal ions with adenine-nucleotides

چکیده
It is well-known that metal ion complexes are essential in various biological systems, including those with adenosine nucleotides which are substrates for a large number of enzymatic processes. The interactions of various metal ions with adenosine nucleotides have been intensively studied by multinuclear NMR spectroscopy. Nucleotides are polydentate ligands with various potential binding sites, including nitrogen atoms on the purine base, hydroxyl groups on the ribose sugar, and negatively charged oxygen atoms in the phosphate group. Depending on the experimental conditions (e.g. pH, concentration range, etc.) and on the size and nature of the metal ions, monodentate, or multidentate coordination to these donor atoms are possible. The review focuses on the applications of different NMR techniques in identifying the stoichiometry and the mode of metal binding in complexes formed with the most important adenosine nucleotides, like adenosine-5′-mono-, diand triphosphates (AMP, ADP and ATP). Ligand exchange dynamics for some metal ion complexes are also presented. © 2008 Elsevier B.V. All rights reserved.

Tetra-Schiff bases derived from (chiral) bis-salphen ligand scaffolds furnish, upon metalation with appropriate metal reagents, their multinuclear structures with associated Zn(OAc)(2) or Zn(OH)(2) fragments. The tendency of retaining these salts was investigated using four different (chiral) bis-salphen scaffolds. The presence of the additional Zn ions was supported by NMR studies, mass determ...

We describe a general multinuclear (1H, 23Na, 87Rb) NMR approach for direct detection of alkali metal ions bound to G-quadruplex DNA. This study is motivated by our recent discovery that alkali metal ions (Na+, K+, Rb+) tightly bound to G-quadruplex DNA are actually "NMR visible" in solution (Wong, A.; Ida, R.; Wu, G. Biochem. Biophys. Res. Commun. 2005, 337, 363). Here solution and solid-state...
متن کاملDynamics and metal ion binding in the U6 RNA intramolecular stem-loop as analyzed by NMR.
The U6 RNA intramolecular stem-loop (ISL) is a conserved component of the spliceosome, and contains an essential metal ion binding site centered between a protonated adenine, A79, and U80. Correlated with protonation of A79, U80 undergoes a base-flipping conformational change accompanied by significant helical movement. We have investigated the dynamics of the U6 ISL by analyzing the power depe...
متن کاملMetal ion-RNA interactions studied via multinuclear NMR.
Metal ions are indispensable for ribonucleic acids (RNAs) folding and activity. First they act as charge neutralization agents, allowing the RNA molecule to attain the complex active three dimensional structure. Second, metal ions are eventually directly involved in function. Nuclear magnetic resonance (NMR) spectroscopy offers several ways to study the RNA-metal ion interactions at an atomic l...
